Safe DNA Gel Stain is a fluorescent nucleic acid dye developed for visual detection of DNA or RNA fragments in agarose or polyacrylamide gels during electrophoresis procedures Structurally this reagent binds nucleic acids by intercalating into their double-stranded regions resulting in enhanced fluorescence upon excitation by ultraviolet (UV) or blue light transilluminators It is commonly utilized in molecular biology workflows such as agarose gel electrophoresis polyacrylamide gel electrophoresis and nucleic acid gel purification Designed as a safer alternative to ethidium bromide it reduces mutagenic risks associated with DNA staining and UV exposure thus facilitating downstream recombinant and cloning experiments Safe DNA Gel Stain is provided as a concentrated solution compatible with DNA and RNA visualization in routine laboratory gel-based analyses

Vector Biolabs AAV-RFP-iCre (AAV1) AAV (AAV1-RFP-iCre)
Cre recombinase is used as a tool to genetically modify genes, such as to delete a segment of DNA flanked by LoxP sites in cells or experimental animals.By applying the mammalian codon usage to Cre recombinase, expression of Cre is improved in the mammalian cells. This improved Cre (iCre) gene also reduce the high CpG content of the prokaryotic coding sequence, thereby reducing the chances of epigenetic silencing in mammals. This AAV1 stock expresses an improved Cre (iCre) along with a RFP under a control of the same CMV promoter. The Cre and RFP was separated by a 2A peptides.

Vector Biolabs cAMP responsive element binding protein 1 Adenovirus (Ad-CREB1)
The ATF/CREB family consists of a series of transcription factors that function by binding to the cAMP responsive element (CRE) palindromic octanucleotide, TGACCTCA. The best characterized members of this gene family include CREB-1, CREB-2, LZIP (also designated CREB3 and Luman), CREM-2, ATF-1, ATF-2, ATF-3, ATF-4, ATF-6 and ATF-7. These transcription factors share terminal leucine zipper dimerization and basic DNA binding domains and are highly variable in their N-termini. Although each of the ATF/CREB proteins bind CREs in their homodimeric form, they can also bind as heterodimers, both within the ATF/CREB family and with members of the AP-1 transcription factor family. Protein kinase A-mediated CREB phosphorylation induces association with a 265 kDa nuclear protein designated CBP (CREB-binding protein), which may represent a CREB coactivator.

Vector Biolabs p16/CDKN2A Adenovirus (Ad-CMV-p16)
p16/CDKN2A is a cyclin-dependent kinase inhibitor that has shown prognostic utility in some human cancers. p16 is frequently mutated or deleted in a wide variety of tumors, and is known to be an important tumor suppressor gene.p16/CDKN2A generates several transcript variants which differ in their first exons. At least three alternatively spliced variants encoding distinct proteins have been reported, two of which encode structurally related isoforms known to function as inhibitors of CDK4 kinase. The remaining transcript includes an alternate first exon located 20 Kb upstream of the remainder of the gene; this transcript contains an alternate open reading frame(ARF) that specifies a protein which is structurally unrelated to the products of the other variants.

Vector Biolabs AAV-DJ-mCherry-iCre AAV (AAVDJ-mCherry-iCre)
Cre recombinase is used as a tool to genetically modify genes, such as to delete a segment of DNA flanked by LoxP sites in cells or experimental animals. By applying the mammalian codon usage to Cre recombinase, expression of Cre is improved in the mammalian cells. This improved Cre (iCre) gene also reduce the high CpG content of the prokaryotic coding sequence, thereby reducing the chances of epigenetic silencing in mammals. The AAV-DJ is a synthetic serotype made from DNA family shuffling of 8 wild type serotypes of AAV, including AAV2, 4, 5, 8, 9, avian, bovine and goat AAV. AAV-DJ outperformed standard AAV serotype 1-8 in cultured cells. This AAV-DJ stock expresses an improved Cre (iCre) and a mCherry driven by one CMV promoter. The iCre and mCherry was separated by a 2A peptides.

Vector Biolabs T-cell factor 4 Adenovirus (Ad-TCF4)
T-cell factor (TCF-1) and lymphoid enhancer-binding factor (for LEF-1, also designated TCF-1?) compose a family of DNA-binding transcriptional activators that are essential for lymphoid cell development. These transcription factors are activated by the Wnt-1 and Wingless pathways and are characterized by the presence of a conserved protein motif, the high mobility group (HMG) 1 box, which mediates DNA binding. LEF-1, TCF-1, and two other family members, TCF-3 and TCF-4, directly bind to cytosolic beta-catenin and facilitate its translocation to the nucleus, where these complexes induce expression of Wnt target genes.

Vector Biolabs GCaMP6m Adenovirus (Ad-GCaMP6m)
GCaMP is a genetically encoded calcium indicator (GECI) which was created from a fusion of green fluorescent protein (GFP), calmodulin, and M13, a peptide sequence from myosin light chain kinase. GECI binds Ca2+ and induce a change in fluorescence signal, which allows measuring action potentials and other receptor activation events that trigger Ca2+ fluxes. The advantage of GECI's are that they can be genetically specified for studies in living organisms. The new novel GCaMP6 have increased ΔF/F0 and faster kinetics compared to previous GCaMP3 and GCaMP5G.This adenovirus expresses GCaMP6m.

Vector Biolabs aryl hydrocarbon receptor nuclear translocator Adenovirus (Ad-HIF1b/ARNT)
The aryl hydrocarbon (Ah) receptor is involved in the induction of several enzymes that participate in xenobiotic metabolism. The ligand-free, cytosolic form of the Ah receptor is complexed to heat shock protein 90. Binding of ligand, which includes dioxin and polycyclic aromatic hydrocarbons, results in translocation of the ligand-binding subunit only to the nucleus. Induction of enzymes involved in xenobiotic metabolism occurs through binding of the ligand-bound Ah receptor to xenobiotic responsive elements in the promoters of genes for these enzymes. This gene encodes a protein that forms a complex with the ligand-bound Ah receptor, and is required for receptor function. The encoded protein has also been identified as the beta subunit of a heterodimeric transcription factor, hypoxia-inducible factor 1 (HIF1)

Vector Biolabs Retinoblastoma-like 2 (p130) Adenovirus (Ad-p130)
The retinoblastoma susceptibility protein Rb is a 110 kDa phosphorylated protein. Like p53, Rb p110 is an anti-oncogene that is subject to inactivation by either mutation or by binding to certain DNA tumor virus-encoded proteins. Rb binds to and regulates transcription factors involved in cell cycle regulation, including the E2F family. Two Rb related proteins, p107 and p130, also function to regulate specific members of the E2F transcription factor family. Binding and inactivation of E2F proteins by Rb is regulated by cyclin dependent kinase-mediated phosphorylation.
